The barcoding protocol within Clavibacter
The genus Clavibacter contains currently six species of which only the species Clavibacter michiganensis is divided into four subspecies. For the identification of the quarantine subspecies C. michiganensis subspecies michiganensis and the quarantine species C. insidiosus and C. sepedonicus, a partial sequence of the Gyrase B gene (GyrB) of approximately 590 bp is amplified using GyrB primers (Richert et al., 2005) with a M13-tag and sequenced using M13 primers.
The protocol for the conventional PCR gyrB for Clavibacter species is available in the annex 2, section 2.3 of PM 7/129.
