The barcoding protocol within Xylella


After 16S rDNA-based identification of the Xylella bacterial genus according to PM7/129, the mutS  barcode can be used to identify Xylella fastidiosa subspecies fastidiosa, multiplex, pauca, sandyi and morus (see also the molecular decision scheme). Please note that strains exist that cannot be unambiguously assigned to a subspecies on the basis of whole genome sequences.

The protocol for the conventional PCR 16S rDNA for bacteria is available in the annex 2, section 2.2 of PM 7/129.
